RESUMO

A yellow-emitting cationic iridium(III) complex [(dfppy)2Ir(TBD)]PF6 (TBD: N4,N4'-bis(3-(triethoxysilyl)propyl)-[2,2'-bipyridine]-4,4'-dicarboxamide; dfppy: 2-(2,4-difluorophenyl)pyridine) containing hydrolysable alkoxysilanes was synthesized. Then, a series of silica-based hybrid nanospheres with diameters of around 400 nm was prepared via the hydrolysis of this complex together with tetraethyl orthosilicate (TEOS, a silica source). When the amount of the complex used was 5.0 wt%, hybrid nanospheres showed the best photoluminescence (PL) properties, relative to the PL quantum yield of pure solid [(dfppy)2Ir(TBD)]PF6 (12.7%), that of hybrid nanospheres increased to 26.2%. Moreover, the thermal decomposition temperature (Td) of pure solid [(dfppy)2Ir(TBD)]PF6 was 331 °C, the Td of the complex in hybrid nanospheres increased to 447 °C. However, the yellow light emission was almost unchanged and was still located at 500-750 nm with a maximum wavelength (λem,max) of 577 nm. Under the excitation of blue-emitting chips (λem,max ≈ 455 nm), cold/neutral/warm white light-emitting diodes (WLEDs) with good luminous quality can all be fabricated using these hybrid nanospheres as phosphors in epoxy resin at different blending concentrations. Compared with two or three iridium(III) complexes being contained in silica-based particles as phosphors as described in literatures, in this study, silica-based hybrid nanospheres covalently containing only one yellow-emitting cationic iridium(III) complex as phosphors provide a more effective and simpler method for preparation high-performance WLEDs.

BACKGROUND: The profound influence of orthodontic treatments on facial aesthetics has been a topic of increasing interest. This study delves into the intricate interplay between orthodontic treatments, facial feature alterations, and aesthetic perceptions. METHODS: A total of 73 patients who had undergone orthodontic treatment were included in this study. Facial photographs were taken before and after treatment. Ten orthodontists provided facial aesthetic ratings (FAR) for each patient's frontal, profile, and overall views. 48 facial landmarks were manually placed by the orthodontists and normalized using Generalized Procrustes analysis (GPA). Two types of phenotypes were derived from facial landmarks. Global facial phenotypes were then extracted using principal component analysis (PCA). Additionally, 37 clinical features related to aesthetics and orthodontics were extracted. The association between facial features and changes in FAR after orthodontic treatment was determined using these two types of phenotypes. RESULTS: The FAR exhibited a high correlation among orthodontic experts, particularly in the profile view. The FAR increased after orthodontic treatment, especially in profile views. Extraction of premolars and orthognathic surgery were found to result in higher FAR change. For global facial phenotypes, the most noticeable changes in the frontal and profile views associated with FAR occurred in the lip area, characterized by inward retraction of the lips and slight chin protrusion in the profile view, as well as a decrease in lip height in the frontal view. The changes observed in the profile view were statistically more significant than those in the frontal view. These facial changes were consistent with the changes from orthodontic treatment. For clinical features, two profile features, namely pg.sm.hori and pg.n.ls, were found to be associated with FAR following orthodontic treatment. The highest FAR scores were achieved when pg.sm.hori was at 80° and pg.n.ls was at 8°. On the other hand, frontal clinical features had a subtle effect on FAR during orthodontic treatment. CONCLUSIONS: This study demonstrated that orthodontic treatment improves facial aesthetics, particularly at lip aera in the profile view. Profile clinical features, such as pg.sm.hori and pg.n.ls, are essential in orthodontic treatment which could increase facial aesthetics.

Rice leaf folder, Cnaphalocrocis medinalis (Guenée), is one of the most serious pests on rice. At present, chemical control is the main method for controlling this pest. However, the indiscriminate use of chemical insecticides has non-target effects and may cause environmental pollution. Besides, leaf curling behavior by C. medinalis may indirectly reduce the efficacy of chemical spray. Therefore, it is crucial to cultivate efficient rice varieties resistant to this pest. Previous studies have found that three different rice varieties, Zhongzao39 (ZZ39), Xiushui134 (XS134), and Yongyou1540 (YY1540), had varying degrees of infestation by C. medinalis. However, it is currently unclear whether the reason for this difference is related to the difference in defense ability of the three rice varieties against the infestation of C. medinalis. To explore this issue, the current study investigated the effects of three rice varieties on the growth performance and food utilization capability of the 4th instar C. medinalis. Further, it elucidated the differences in defense responses among different rice varieties based on the differences in leaf physiological and biochemical indicators and their impact on population occurrence. The results showed that the larval survival rate was the lowest, and the development period was significantly prolonged after feeding on YY1540. This was not related to the differences in leaf wax, pigments, and nutritional components among the three rice varieties nor to the feeding preferences of the larvae. The rate of superoxide anion production, hydrogen peroxide content, and the activity of three protective enzymes were negatively correlated with larval survival rate, and they all showed the highest in YY1540 leaves. Compared to other tested varieties, although the larvae feeding on YY1540 had higher conversion efficiency of ingested food and lower relative consumption rate, their relative growth was faster, indicating stronger food utilization capability. However, they had a lower accumulation of protein. This suggests that different rice varieties had different levels of oxidative stress after infestation by C. medinalis. The defense response of YY1540 was more intense, which was not conducive to the development of the larvae population. These results will provide new insights into the interaction mechanism between different rice varieties and C. medinalis and provide a theoretical basis for cultivating rice varieties resistant to this pest.

The residues of pesticides and antibiotics have always been a major concern in agriculture and food safety. In order to provide a new method for the rapid detection of organophosphorus pesticides and antibiotics, a novel Cd(II) chain-based zwitterionic metal-organic framework MOF 1 with high sensitivity fluorescence sensing performance was successfully synthesized. A series of researches showed that the water- and pH-stable bifunctional MOF 1 has a great ability to detect phosmet (PSM) and chlortetracycline (CTC) in water through fluorescence quenching effect, with high detection sensitivity, low detection limits (0.0124 µM and 0.0131 µM), short response time (40 s) and reusability. Practical application results revealed that MOF 1 could detect PSM and CTC in milk, beef, chicken and egg samples, with satisfactory recoveries (95.2%-103.7%). As a novel fluorescence probe, MOF 1, is known the first case that can detect PSM in animal-derived samples, and the first dual-function material capable of detecting PSM and CTC. Mechanism studies displayed that competitive absorption and photoinduced electron transfer clearly authenticate the high quenching performance of the material.

The advancement in CRISPR-Cas biosensors has transmuted the detection of plant viruses owing to their rapid and higher sensitivity. However, false positives and restricted multiplexing capabilities are still the challenges faced by this technology, demanding the exploration of novel methodologies. In this study, a novel detection system was developed by integrating reverse transcriptome (RT) techniques with recombinase polymerase isothermal amplification (RPA) and Pyrococcus furiosus Argonaute (PfAgo). The RT-RPA-PfAgo system enabled the simultaneous detection of rice ragged stunt virus (RRSV), rice grassy stunt virus (RGSV), and rice black streaked dwarf virus (RBSDV). Identifying targets via guide DNA without being hindered by protospacer adjacent motif sequences is the inherent merit of PfAgo, with the additional advantage of it being simple, cost-effective, and exceptionally sensitive, with detection limits between 3.13 and 5.13 copies/µL, in addition to it effectively differentiating between the three distinct viruses. The field evaluations were also in accordance with RT-PCR methods. The RT-RPA-PfAgo system proved to be a robust, versatile, highly specific, and sensitive method with great potential for practicality in future plant virus diagnostics.

The brown planthopper (BPH), Nilaparvata lugens, is one of the most destructive pests of rice. Given the threats posed by insecticide resistance to its control, eco-friendly strategies based on microbial pathogens emerged as a promising biocontrol alternative. In the present study, we isolated a native fungal pathogen against BPH from infected BPH cadavers and preliminarily identified as a strain of Aspergillus fumigatus based on morphological and molecular methods. Laboratory bioassay revealed that this fungal strain was highly virulent to BPH both at nymphal and adult stages, with the median lethal times (LT50) of 7.5 and 5.8 days under high conidial concentration of 1 × 109 conidia mL-1. A genome-wide view of gene expressions in BPH against fungal attack was analyzed by transcriptomic sequencing and consequently a large number of differentially expressed genes that mainly involved in host immune defense and cell detoxification were found. RNAi-mediated knockdown of an upregulated gene encoding a serine protease (NlSPN) could cause a significant decrease in BPH survival. Combination of dsRNA injection and fungal infection showed an additive effect on BPH mortality, which provided clues to develop new pest management strategies against BPH.

Because rare-earth elements are scarce, expensive, and unsustainable, it is of great significance to develop rare-earth-free (even metal-free) luminescent materials as phosphors for LEDs. Here, a graphitic-C3N4 (g-C3N4) derivative containing some heptazines merged with phenyls has been synthesized via thermal polymerization of melamine and quinazoline-2,4(1H,3H)-dione at an optimal mole ratio of 18 : 1. In comparison with g-C3N4 synthesized from melamine only, the photoluminescent (PL) emission colour changed from blue to green, the maximum emission wavelength (λ em,max) changed from 467 nm to 508 nm, and the PL quantum yield (PLQY) increased from 8.0% to 24.0%. It was further purified via vacuum sublimation, and a product with yellowish green emission (λ em,max = 517 nm) and PLQY up to 45.5% was obtained. This sublimated product had high thermal stability and low thermal quenching; its thermal decomposition temperature was as high as 527 °C, and its relative PL emission intensity at 100 °C was 90.8% of that at 20 °C. Excited by blue light chips (λ em,max ≈ 460 nm), cold, neutral and warm white LEDs can be fabricated using the sublimated product and orange-emitting (Sr,Ba)3SiO5:Eu2+ as phosphors. The good performances of these white LEDs (for example, the CIE coordinates, color rendering index and correlated color temperature were (0.31, 0.33), 84.4 and 6577 K, respectively) suggest that the low-efficiency blue-emitting g-C3N4 had been successfully converted into a high-efficiency metal-free quasi-green phosphor.

Fluorescent probes always attract huge attention. In particular, carbon dots, due to their unique biocompatibility and variable fluorescence characteristics, can be used in multiple fields and were fulled of expectation by researchers. Since the advent of the dual-mode carbon dots probe, which greatly improved the accuracy of quantitative detection, there are higher hopes for dual-mode carbon dots probes. Herein, we have successfully developed a new dual-mode fluorescent carbon dots probe based on 1,10-phenanthroline (Ph-CDs). Ph-CDs detect the object to be measured based on both down-conversion luminescence and up-conversion luminescence at the same time, different from the reported dual-mode fluorescent probes which are based on the wavelength and intensity changes in down-conversion luminescence. As-prepared Ph-CDs have good linear relationships with the polarity of solvents in down-conversion luminescence (R2 = 0.9909) and up-conversion luminescence (R2 = 0.9374), respectively. Hence, Ph-CDs provide a new in-depth insight into the design of fluorescent probes with dual-mode detection and they can give more accurate, reliable and convenient detection results.

Low-cost fish species are often used to adulterate or substitute for Atlantic salmon products, posing a serious threat to market order and public health. Hence, reliable techniques are urgently needed to detect Atlantic salmon adulteration. In this study, a precise method for identifying and quantifying adulterated Atlantic salmon with rainbow trout based on droplet digital PCR (ddPCR) testing was developed. Species-specific primers and probes were designed targeting the single-copy nuclear gene myoglobin of two salmonids. A quantitative formula for calculating the mass fraction of adulterated Atlantic salmon with rainbow trout was established based on a one-step conversion strategy, in which the DNA copy number ratios were directly transformed to meat mass fractions by introducing a fixed constant (the transfer coefficient). The dynamic range of the established ddPCR method was from 1% to 90%, with a limit of detection (LOD) of 0.2% and a limit of quantification (LOQ) of 0.8% for rainbow trout in Atlantic salmon, respectively. The quantification method demonstrated an acceptable level of repeatability and reproducibility, as the values of the relative standard deviation (RSD) for the tested meat mixtures with the known fractions were all less than 5%. Thermal and freezing treatments, as well as adding food additives within the recommended dosage limits, had no significant effect on the quantification accuracy. The method was successfully applied to detect rainbow trout adulteration in commercial raw and processed Atlantic salmon products. In comparison to real-time quantitative PCR (qPCR) testing, the established ddPCR method exhibited a higher level of stability and accuracy. Overall, the ddPCR-based quantitative method exhibited high levels of accuracy, stability, sensitivity, and practicability, suitable for applications in the routine surveillance and quality assurance of salmon products.

Cnaphalocrocis medinalis is a destructive migratory rice pest. Although many studies have investigated its behavioral and physiological responses to environmental changes and migration-inducing factors, little is known about its molecular mechanisms. This study was conducted to select suitable RT-qPCR reference genes to facilitate future gene expression studies. Here, thirteen candidate housekeeping genes (EF1α, AK, EF1ß, GAPDH, PGK, RPL13, RPL18, RPS3, 18S rRNA, TBP1, TBP2, ACT, and UCCR) were selected to evaluate their stabilities under different conditions using the ∆CT method; the geNorm, NormFinder, BestKeeper algorithms; and the online tool RefFinder. The results showed that the most stable reference genes were EF1ß, PGK, and RPL18, related to developmental stages; RPS3 and RPL18 in larval tissues; EF1ß and PGK in larvae feeding on different rice varieties; EF1α, EF1ß, and PGK in larvae temperature treatments; PGK and RPL13, related to different adult ages; PGK, EF1α, and ACT, related to adult nutritional conditions; RPL18 and PGK, related to adult mating status; and, RPS3 and PGK, related to different adult take-off characteristics. Our results reveal reference genes that apply to various experimental conditions and will greatly improve the reliability of RT-qPCR analysis for the further study of gene function in this pest.

In this work, a series of Cr3+-doped A2NaYF6 (denoted as ANYF:Cr; A = K, Rb, and Cs) fluoroyttrate double-perovskites with broad blue excitation and near-infrared (NIR) emission bands were prepared using a high-temperature solid-state reaction method. The physicochemical and spectroscopic properties of the as-prepared products, including crystal structure, bandgap, morphology, luminescence behaviour and optical performance, were studied in detail. The results show that the Cr3+ activators undergo a decreasing crystal field in ANYF in the direction from K to Cs, yielding a red shift of the emission peak position from 758 to 786 nm, whilst the full-width-at-half-maximum (FWHM) expands from 98 to 104 nm. By virtue of the broadband NIR emission, clear structural images of the tangerine section, mini LED flashlight and circuit board were obtained by merging the optimal RNYF:Cr phosphor on a blue InGaN chip, implying its potential use in light-emitting diodes for non-destructive testing.

The brown planthopper (Nilaparvata lugens, BPH) is a destructive insect pest posing a serious threat to rice production. The fungal entomopathogen Metarhizium anisopliae is a promising alternative that can be used for BPH biocontrol. Recent studies have highlighted the significant involvement of gut microbiota in the insect-fungus interactions. In the presented study, we investigated the effects of topical fungal infection on the gut microbial community structure in BPH. Our results revealed that topical infection with M. anisopliae increased the bacterial load and altered the bacterial community structure in the gut of BPH. The relative abundances of the dominant gut bacteria at the order, family and genus level were significantly different between fungus-infected and uninfected groups. At the genus level, the uninfected BPH harbored high proportions of Pantoea and Enterobacter in the gut, whereas the fungus-infected BPH gut was absolutely dominated by Acinetobacter. Moreover, topical fungal infection significantly inhibited the expressions of immune-related genes encoding anti-microbial protein and dual oxidase that were involved in the maintenance of gut microbiota homeostasis, indicating that gut bacteria imbalance might be attributed in part to the suppression of gut immunity caused by fungal pathogen. Our results highlighted the importance of the gut microbial community during interactions between fungal pathogens and insect hosts.

Luminescent crystals with high efficiency have huge potential in applications for white light-emitting diodes (LEDs). Herein, organic-inorganic hybrid crystals doped with Mn4+, [N(CH3)4]2XF6:Mn4+ (X = Si, Ge, and Ti), were grown under mild conditions. Their crystal structural evolution was confirmed by single-crystal X-ray diffraction at different temperatures. These crystals exhibit intense red emission with a high external quantum efficiency (73.0% for [N(CH3)4]2TiF6:Mn4+) and good thermal stability. The warm white LEDs were fabricated by combining these red-emitting phosphors with a YAG:Ce3+ ceramic chip. As-grown crystals can significantly optimize the performances of white LEDs (colour rendering index up to 95). Hence, this work provides a new strategy to explore Mn4+-activated organic-inorganic hybrid materials for white LEDs.

The non-equivalent doping of Mn4+ in red-emitting fluoride phosphors effectively shortens the fluorescence lifetime. Herein, we successfully synthesized Rb2NaInF6:Mn4+ phosphors by an ion-exchange method. The compensation mechanism of Mn4+ local symmetry and charge balance in the phosphor were studied in detail by theoretical calculations based on density functional theory. The phosphor Rb2NaInF6:Mn4+ (5.59 mol%) exhibits intense red emission with a short fluorescence lifetime (1.45 ms). Meanwhile, it has excellent colour stability and thermal quenching stability at elevated temperatures. Using Rb2NaInF6:Mn4+, ß-SiAlON:Eu2+ and a blue-emitting GaN chip as red, green, and blue components, the white light-emitting diode (w-LED) exhibits a wide colour gamut of 94.1% of the National Television System Committee (NTSC) standard value. So, this work provides valuable information to investigate novel red-emitting fluoride phosphors for LED backlighting.

Adulteration of high-value beef with lower-priced alternatives is a world-wide problem resulting in consumers' distrust and market chaos. Therefore, effective methods for the identification and quantification of adulterated beef products are urgently needed. In this study, we developed a reliable droplet digital PCR (ddPCR) method targeting the single-copy nuclear genes for qualitative and quantitative detection of the presence of porcine and chicken derivatives in beef. A fixed constant (transfer coefficient) was introduced to directly transform the ratio of DNA copy number to the mass proportion of targeted meats. Results revealed that the linearity range of quantification for pork and chicken were both from 1% (w/w) to 90% (w/w). The limit of detection (LOD) and limit of quantification (LOQ) of the developed ddPCR method were the same for pork and chicken in beef, with LOD 0.1% (w/w) and LOQ 1% (w/w). The accuracy and applicability of the method was tested and verified using mixed samples with the known proportions and commercially available beef products. We conclude that our developed ddPCR method was accurate and reliable in identifying and quantifying porcine and chicken derivatives in beef and therefore has great potential to be applied in routine analyses and quality control of beef products.

The zero-phonon line (ZPL) of Mn4+, which highly depends on its local environment, is usually much weaker than the vibrational phonon sidebands. In this work, an ultraintense ZPL emission, coming from a brand new red-emitting Rb2LiGaF6:Mn4+ (RLGFM) phosphor upon blue light excitation, is presented. The interesting spectral characteristic originates from the nonequivalent substitution of Mn4+ for Ga3+ in a rigid octahedral structure with a low symmetry, which induces neighboring cation vacancies that distort the local symmetry of the [MnF6] octahedra. Benefiting from the ultraintense ZPL emission, a wide color gamut is achieved using RLGFM and ß-SiAlON:Eu2+ as color converters. Moreover, a comprehensive investigation on the thermal quenching behavior is also conducted to provide detailed insights to explore novel Mn4+ red phosphors for high-quality backlight display applications.

In this work, we present a brand-new narrowband red-emitting fluoroperovskite via the introduction of Mn4+ into NaZnF3 through a facile co-precipitation method at room temperature. The physicochemical properties of the fluoroperovskite such as crystal and electronic structures, morphology, and elemental composition, as well as its spectroscopic properties such as luminescence behaviours and optical performance were characterized and investigated in detail. Evidence shows that NaZnF3:Mn4+ exhibits a uniform particulate shape with single-phase crystallinity. By virtue of the non-equivalent substitution and the [MnF6] octahedral distortion in the fluoride host, sharp red emissions of phonon sidebands and the zero-phonon line upon blue light excitation are identified. Benefiting from the unique spectral feature, a wide colour gamut of 104.1% NTSC is achieved by coating ß-SiAlON:Eu2+ and NaZnF3:Mn4+ on an InGaN chip, indicating the potential use of the Mn4+ fluoroperovskite as a colour converter for display backlight application.

BACKGROUND: Timely and appropriate administration of post-exposure prophylaxis (PEP) is an essential component of human rabies prevention programs. We evaluated patient care at rabies clinics in a high-risk county in Hunan Province, China to inform strategies needed to achieve dog-mediated human rabies elimination by 2030. METHODS: We collected information on PEP, staff capacity, and service availability at the 17 rabies clinics in the high-risk county during onsite visits and key staff interviews. Additionally, we conducted observational assessments at five of these clinics, identified through purposive sampling to capture real-time information on patient care during a four-week period. Wound categories assigned by trained observers were considered accurate per national guidelines for comparison purposes. We used the kappa statistic and an alpha level of 0.05 to assess agreement between observers and clinic staff. RESULTS: In 2015, the 17 clinics provided PEP to 5,261 patients. Although rabies vaccines were available at all 17 clinics, rabies immune globulin (RIG) was only available at the single urban clinic in the county. During the assessment period in 2016, 196 patients sought care for possible rabies virus exposures. According to observers, 88 (44%) patients had category III wounds, 104 (53%) had category II wounds and 4 (2%) had category I wounds. Observers and PEP clinic staff agreed on approximately half of the assigned wound categories (kappa = 0.55, p-value< 0.001). Agreement for the urban county-level CDC clinic (kappa = 0.93, p-value<0.001) was higher than for the township clinics (kappa = 0.16, p-value = 0.007). Using observer assigned wound categories, 142 (73%) patients received rabies vaccinations and RIG as outlined in the national guidelines. CONCLUSION: Rabies PEP services were available at each town of the project county; however, gaps between clinical practice and national rabies guidelines on the use of PEP were identified. We used these findings to develop and implement a training to rabies clinic staff on wound categorization, wound care, and appropriate use of PEP. Additional risk-based approaches for evaluating human rabies virus exposures may be needed as China progresses towards elimination.

BACKGROUND: The brown planthopper (BPH) is one of the most destructive pests of rice, causing tremendous yield and economic losses every year. The fungal entomopathogen Metarhizium anisopliae was previously proved to have great potential for BPH biocontrol. Genome-wide insight into the insect-fungus interaction is crucial for genetic improvement of M. anisopliae to enhance its virulence to BPH but still has been poorly explored. RESULTS: Using dual RNA-seq approach, we present here a global view of host and fungal gene expressions in BPH adults during the fungal infection. The results revealed that BPH could initiate strong defense responses against the fungal attack by upregulating the expressions of a large number of genes, including genes involved in cuticle formation, immune response, cell detoxification and biomacromolecule metabolism. Correspondingly, the fungal entomopathogen could induce a series of genes to infect and modulate BPH, including genes involved in fungal penetration, invasive growth, stress resistance and virulence. Three host defense-related genes (NlPCE4, NlPOD1 and NlCYP4DE1) were chosen for further function analysis. RNAi-mediated knockdown of NlPCE4 caused a significant decrease in BPH survival, but no obvious effects on the survival rates were detected by the suppression of NlPOD1 and NlCYP4DE1. Combination of dsRNA injection and fungal infection could significantly enhance the BPH-killing speed, as synergistic mortalities were observed in co-treatments of RNAi and M. anisopliae infection. CONCLUSION: Our study provides a comprehensive insight into molecular mechanisms of host-pathogen interaction between BPH and M. anisopliae and contributes to future development of new efficient biocontrol strategies for BPH biocontrol.

Microbiome associated with insects play vital roles in host ecology and physiology. The small brown planthopper (SBPH), Laodelphax striatellus, is a polyphagous insect pest that caused enormous damage to a wide range of cereal crops. Previous studies have assessed the effects of environmental factors, such as antibiotics, insecticide, and geographical habitat on the bacterial composition of SBPH. However, the influence of host plants on the microbial community in SBPH still unclear. Here, we characterized and compared the microbial community in three SBPH populations feeding on rice, barley, and wheat, respectively, using high-throughput amplicon sequencing. Our observations revealed that the microbiome harbored by SBPH was abundant and diverse. Ten phyla comprising 141 genera of bacteria were annotated, and four fungal phyla consisting of 47 genera were assigned. The bacteria belonging to the phylum Proteobacteria were the most prevalent and the fungi with the highest abundance were from the order Hypocreales. Comparative analysis showed that host plants could significantly induce structural changes of SBPH microbiome. Significant differences in abundance were observed in two main bacterial orders (Rickettsiales and Rhodospirillales) and three fungal classes (Sordariomycetes, an unclassified class in Ascomycota and Eurotiomycetes) among three host-adapted SBPH populations. Our results could broaden our understanding of interactions among SBPH, its microbial associates and host plants, and also represented the basis of future SBPH biological management.
